Simple question regarding member upgrade

If a member is manually upgraded will PayPal update automatically to the higher amount next pay period via IPN?

Simple answer: No

Existing subscription contracts will continue at the same rate. And PayPal IPN will continue to be be sent, but only to indicate payment, payment failure, or cancellation of the previously existing subscription contract.

Another question, can I upgrade the amount from the website? Or do they need to cancel and sign up again?

For an existing subscription, it needs to be cancelled (since PayPal won’t let you change the amount of an existing subscription to prevent any kind of fraud). Then you can request the member to pay using the new payment button which will have the new subscription amount.
